Early work: ABS-CBN (1992–1997)

Barretto was discovered by talent manager Douglas Quijano in 1992 through older sister,Gretchen Barretto,also an actress. That same year, she landed her first major role on television when she was cast forABS-CBN's youth-oriented variety show,Ang TV.

In 1993, she starred at television seriesHome Along Da RileswithDolphywhich further introduced her to local media. She also appeared in television shows likeOki Doki Doc(1993–2000),Palibhasa Lalake(1993–1998) and movieMay Minamahal(1993).

In 1994, she starred inPare KowithMark Anthony Fernandezand inOki Doki Doc: The Movie.

In 1996, she starred in various films withStar Cinemawhich includes,Radio Romance,her first film withRico Yan,Mangarap Ka,her first major film role withMark Anthony Fernandez,andMadrastawithChristopher de LeonandSharon Cuneta.

Barretto landed her first ever television drama series in 1997 titledMula sa Puso.The series ran for three consecutive years onABS-CBN.That same year she starred in films likeHome Along Da Riles 2withDolphy,Calvento Files: The MoviewithRio LocsinandDiether Ocampo,andF.L.A.M.E.S.: The MoviewithRico Yan,Paula Peralejo,andBoots Anson-Roa.

In 1998, she portrayed Mela in the filmDahil Mahal na Mahal Kitawhich was directed byWenn V. Deramas,alongsideRico YanandDiether Ocampo.

Later in 1999, with the debut of the soap operaSaan Ka Man Naroroonwhere she portrayed astripletsin the name of Rosario, Rosenda and Rosemarie which earned her a nomination for Best Actress in theAsian TV Awards.Also the same year she starred in films likeSolterawithDiether OcampoandMaricel Soriano,and the film adaptation ofMula sa Puso.

She also co-starredVilma Santosin the highly commended 2000 movieAnakdirected byRory Quintos.

Move to GMA Network (2009–2012)

Barretto shocked the local media in November 2009 when she broke ties with her long-time networkABS-CBNand transferred to rival companyGMA Network.[1]

In 2010, she started her own weekly drama anthology,ClaudinewithGMA Network.Also that same year she was a part of a movie produced byGMA FilmsandViva FilmstitledIn Your Eyesco-starringAnne CurtisandRichard Gutierrez.

Comeback, TV5 (2015–2017)

Barretto made a major big screen comeback in 2015 starring in theStar CinemamovieEtiquette for MistressesalongsideKim Chiu,Iza Calzado,Cheena Crab,Kris Aquinoand directed byChito S. Roño.Despite the controversy of the film, it was acclaimed critically and was a box-office hit. Claudine also marks her first appearance onMaalaala Mo Kayaafter more than 20 years, in an episode titled"Itak/Bolo"which aired in October of the same year.

In 2016, she joinedTV5and starred in the television drama seriesBakit Manipis ang Ulap?which was directed byJoel Lamanganand co-starringDiether Ocampo,Meg Imperial,andCesar Montano.That same year she appeared in anotherMMKepisode titled"Luneta Park"which aired in December.

In 2017, she made her first ever appearance at theStar Magic Ball,since she leftABS-CBNin 2009.

In 2020, she signed a management contract withALV Talent Circuit.

Second comeback: GMA Network (2017-present)

In 2022, she made a television comeback onGMA Networkthrough leading appearance inWish Ko Lang.She led two episode of the said show for anniversary which both won in television ratings.

In March 2002, a month after the box-office success of their movieGot 2 Believe,Barretto's on-screen and real-life partner of four years,Rico Yan,died of acute hemorrhagic pancreatitis.[2]

Barretto married actorRaymart Santiagoon March 27, 2006, but annulment proceedings had been initiated in 2013.[3]In 2015, she adopted three children, and has one child with Santiago.[4]

On October 7, 2021, Claudine formalized her bid to run as a city councilor forOlongapo Cityin the2022 electionsunder thePDP–Labanpolitical party.[5]On April 12, 2022, Claudine expressed her support for presidential candidateBongbong Marcosin the elections.[6]
